Responsibilities

General Purpose (See percentages below)

Responsible for carrying out computer analyses of development of myelin pathways related to cortical and basal ganglia circuits. This requires performance of multiple techniques including scanning microscope slides, adjusting images, and analyzing data using complex software programs. Other tasks may include some laboratory skills such as, preparing microscope slides, and mounting tissue sections onto slides. Good knowledge of basic computer programing related to biological analyses are required. Willingness to work with animal tissue and the ability to work in a laboratory team environment with a diverse group of personnel is essential.

Typical Assignments and Responsibilities (See percentages below)

ESSENTIAL TASKS

Specific Responsibilities:

20% Divide the frontal cortex for each subject into cortical areas. Each cortical region is divided into subareas based on cortical atlas divisions. Draws sample strips from each cortical area.

20% Perform analysis using a combination of software programs to determine the myelin optical density. Each image is converted to grey scale. Each cortical strip is divided into deep and superficial cortical layers. Using NIH ImageJ optical density is taken for deep and superficial cortical layers for each strip.

20% Run statistical analysis to compare the development of cortical myelin. Analyze the myelin development across ages, across cortical areas and layers. Present findings to PI and/or in laboratory meetings.

15% Prepare sections for analyses. Work in an independent manner with minimal supervision. Select, clean and prepare slides for scanning. Scan slides and convert each image into a separate a JPEG 2000 file. Upload large JPEG 2000 files into the cloud server.

10% Align myelin-stained slides with NeuN-stained tiffs files. Using adjacent sections, with different histological stains, tiff images are aligned based on anatomical landmarks with photoshop.

10% General database maintenance. Provide support and technical assistance on experimental problems that may arise during the course of the experiments. Order supplies related to computer analysis. Maintain functionality of lab databases, including injection sites, and references. Perform literature searches, assist with grant and manuscript preparation, and make scientific contributions to research projects. Other miscellaneous laboratory-related duties assigned as required for research projects.
